These and other corisiderations have prompted us to investigate the possible regulatory roles of distant DNA sequences in relation to genes of the ~-globin cluster and in this communication we summarise some findings which show potential regulatory function for sequences upstream of the human E-globin gene. We initially approached this problem by searching for minor transcription initiation sites at locations upstream of the canonical mRNA cap site of the human E-globin gene. , 1981) and we reasoned that if this was true, such alternative initiation sites might serve as landmarks for distant regions involved in control of the gene.
